The obesity epidemic continues to worsen worldwide, driving metabolic and chronic inflammatory diseases. Thiazolidinediones (TZDs), such as rosiglitazone (Rosi), are PPARy ligands that promote anti-inflammatory changes in adipose tissue macrophages (ATMs) and cause insulin sensitization. We performed single-cell RNA-seuqnecing (scRNA-seq) to analyze the diversity and the phenotype of ATM subpopulations under Rosi and ATM-Exosome treatment. Overall design: We performed a single-cell transcriptome analysis of stromal vascular cells of the epididymal fat isolated from obese mice treated for the last month with Rosiglitazone or with ATM-Exosomes or PBS Ctrl liposomes. Obese mice were fed with HFD for 10 weeks and were then treated for 4 weeks with Rosi-ATM-Exosomes (Rosi-ATM-Exos, n=3), HFD-ATM-Exos (n=4), or PBS Ctrl liposomes (n=3) (10^9 Exos twice a week) compared to Rosi diet (HFD containing 3 mg/kg/day Rosiglitazone, n=3).

Construction protocol: Epididymal fat was isolated, enzymatically digested in a collagen solution, and after red blood cells were lyzed and a cell vaibility of 80-90% confirmed, stromal vascular cells were used for scRNA-seqencing. Cell suspensions from each mouse were loaded into a single well of a 10× Genomics Chromium Single Cell Controller.
