As part of a study on the role of Rap1 at the promoters of HML and MAT in S cerevisiae, we endogenously tagged Rap1 with a 3xV5 tag and performed ChIPseq in a variety of genetic backgrounds including: silenced (SIR); unsilenced (sir1?/sir3?/sir4?); in strains witha wild type alpha2 promoter (wtpromoter); or with a point mutation at the promoter (mutpromoter). Overall design: 3xV5-Rap1 ChIPseq. Replicates are included for every sample.

Construction protocol: Cells were lysed by mechanical disruption in a bead beater. Chromatin was fragmented by sonication. All immunoprecipitations were performed using 50uL DynaBeads Protein G (ThermoFisher Scientific 10003D) magnetic beads coupled with 5uL monoclonal mouse-anti-V5 antibody (ThermoFisher Scientific R96025) DNA was purified using Qiagen spin column kit NEB Ultra II library prep kit
