Study: Transcriptomic changes in embryonic hearts from Dp3Tyb and Ts1Rhr mouse models of Down Syndrome

We performed bulk RNAseq on whole hearts from E13.5 Dp3Tyb embryos and from wild-type littermates, as well as hearts from E13.5 Ts1Rhr embryos and wild-type littermates. Analysis showed the expected increased expression of the Hsa21-orthologous genes on Mmu16 that are present in 3 copies. Gene set enrichment analysis identified pathways that are altered in Dp3Tyb and Ts1Rhr hearts. Overall design: RNAseq on 5 E13.5 Dp3Tyb embryonic hearts and 5 wild-type littermate embryonic hearts, and on 5 E13.5 Ts1Rhr embryonic hearts and 5 wild-type littermate embryonic hearts.

Construction protocol: Hearts were dissected and snap frozen and kept at -80C. Samples were lysed (buffer RLT Qiagen RNeasy Minikit) and homogenised using a battery-operated pestle motor mixer followed by RNA extraction using Qiagen RNeasy Mini kit using the standard protocol. cDNA library synthesis using KAPA mRNA HyperPrep Kit (Illumina) and libraries were sequenced on the Illumina HiSeq 4000 following manufacturer's protocols.
