Study: Induction of mouse totipotent stem cells by a defined chemical cocktail [RRBS]

We demonstrate induction and long-term maintenance of totipotent stem cells (TotiSCs) from mouse pluripotent stem cells (PSCs) by a combination of three small molecules, TTNPB, 1-Azakenpaullone, and WS6. These cells, which we designated as ciTotiSCs (chemically induced totipotent stem cells), resembled mouse totipotent 2C-embryo stage cells at both transcriptome and epigenome level. Overall design: RRBS-seq of ciTotiSC with two replicates.

Construction protocol: Genomic DNAs were extracted using DNeasy Blood and Tissue Kits (QIAGEN) and digested by MspI (NEB). The digested DNAs were filled in and tailed. Then, bisulfate conversion was performed using MethyCode bisulfite conversion kit (Invitrogen). After amplification and purification,all libraries were constructed and subjected to 150 bp paired-ends Illumina Novaseq 6000 platform.
