Study: Adaptive aneuploidy protects against thiol peroxidase deficiency by increasing respiration via key mitochondrial proteins
show Abstracthide Abstract
We characterized the mechanisms that allow yeast cells to survive under conditions of thiol peroxidase deficiency. Two thiol peroxidase null yeast (delta8) strains independently acquired a second copy of chromosome XI and increased expression of genes encoded by it. Delta-8 strains were compared to the Wt-M11 strain, which is characterized by duplication of chr-XI. Overall design: Examination of thiol peroxidase-deficient yeast strains.

Library:
Instrument: Illumina HiSeq 2000
Strategy: RNA-Seq
Source: TRANSCRIPTOMIC
Selection: cDNA
Layout: SINGLE
Construction protocol: Frozen aliquots were thawed and lysed in 400 μL of lysis buffer (mRNA DIRECT kit; Invitrogen). A 250-μL aliquot of magnetic beads and two rounds of purification were implemented according to the manufacturer’s protocol RNA libraries were prepared for sequencing using standard Illumina protocols
