Mice rendered obese with high fat diet were treated by IP injection with N-trans caffeoyltyramine (NCT) (200 mg/kg bid) for 14 days, followed by harvesting of the liver for analysis, including RNA isolation. Overall design: High fat diet induced non-alcoholic fatty liver (NAFLD) treated with DMSO or NCT compound.

Construction protocol: Liver from DMSO or NCT treated mice was removed, flash frozen in liquid nitrogen, and RNA harvested using Trizol reagent. PolyA RNA was isolated using the NEBNext® Poly(A) mRNA Magnetic Isolation Module and barcoded libraries were made using the NEBNext® Ultra II™ Directional RNA Library Prep Kit for Illumina®(NEB, Ipswich MA). Libraries were pooled and single end sequenced (1X75) on the Illumina NextSeq 500 using the High output V2 kit (Illumina Inc., San Diego CA).
