Study: Methylome analysis by reduced representation bisulfite sequencing (RRBS) from adipose tissue samples of piglets from a swine model of maternal overfeeding and/or restriction

Maternal overfeeding and/or restriction may have effect on the metabolic and epigenetic programming of offspring's adipose tissue. The study tested the potential of postnatal metformin teatment to reverse these changes using a swine animal model. Overall design: We designed a swine model using pregnant sows distributed in 3 groups of diet intervention (control, overfed and restricted) through pregnancy. At birth, piglets underwent standard lactation without maternal diet intervention. Within each maternal group, piglets were rambdomly assigned to two different tratment interventions: metformin or vehicle. All piglets were sacrified at weaning and visceral adipose tissue samples were collected.

Construction protocol: High quality genomic DNA (400ng) was isolated from adipose tissue samples with Gentra PureGene tissue kit (Qiagen) according to manufacturer's instructions. DNA was bisulfited with the EZ DNA Methylation-Gold Kit (Zymo Research), according to manufacturer's instructions. Bisulfite conversion rate was over 99%. Libraries were were prepared for sequencing using standard Illumina protocols and Sequencing was performed on a HiSeq 3000 (Illumina).
