This study seeks to assess the effects of prenatal exposure of female sheep to execessive testosterone in metabolically relevant liver and muscle tissue. The goals are to 1) determine genes and gene pathways in the control animals and 2) assess the effects of prenatal T-treatment on gene expression and gene pathways. 3) Finally identify putative ncRNA-totalRNA interactions. Overall design: 10 Female Sheep. 5 Controls and 5 Treatment. 2 Tissues-Liver and Muscle. 9 Samples Liver(4C, 5T) 10 Samples Muscle(5C, 5T)

Construction protocol: Total RNA was extracted from the liver and muscle after homogenizing the tissues in Trizol reagent (Life Technologies, Carlsbad, CA) following the manufacturer's recommendations. Any residual contaminating DNA was removed by binding the RNA in a RNeasy binding column, treating the column with RNAse free DNAse (Qiagen, Germantown, MD) and eluting the RNA in nuclease free water. The quality of purified RNA including the purity and RNA integrity were evaluated using Agilent 2100 bioanalyzer (Agilent Technologies, Santa Clara, CA) at the University of Michigan Advanced Genomics Core. library preparation was carried out using TruSeq RNA Library Prep Kit (Illumina, San Diego, CA)
