Study: Genome-wide identification and profiling of microRNAs in Paulownia tomentosa cambial tissues in response to seasonal changes
show Abstracthide Abstract
Analysis of Paulownia tomentosa cambial tissues at gene expression level. The hypothesis tested in the present study was that miRNAs can regulate the development of the vascular cambium. The results provide new insights into the important regulatory functions of miRNAs in vascular cambium development and wood formation in conifers. Overall design: Examination of 3 different periods in Paulownia tomentosa cambial tissues
Sample: P. tomentosa reactivating stage rep1

Library:
Instrument: Illumina HiSeq 2000
Strategy: RNA-Seq
Source: TRANSCRIPTOMIC
Selection: cDNA
Layout: SINGLE
Construction protocol: total RNAs from dormant, reactivating, and active cambium meristem were isolated using the Trizol reagent The library was constructed using Small RNA Sample Pre Kit. Based on the 3 'and 5' special structures of Small RNA (complete phosphate groups on the 5 'end and hydroxyl groups on the 3' end), total RNA was used as a starting sample and Small Both ends of the RNA were spliced and cDNA was reverse transcribed. Followed by PCR amplification, PAGE gel electrophoresis separation of the target DNA fragment, gel recovery obtained is the cDNA library
