SRX2354038: RNA-seq of Oxytricha trifallax: encysted cells
1 ILLUMINA (Illumina HiSeq 2500) run: 47.8M spots, 3.2G bases, 1.5Gb downloads

Design: We collected vegetatively-growing Oxytricha trifallax JRB310 cells and incubated them, without food, for five days to trigger cell encystment. Cells were collected and lysed with a Nucleospin Tissue Kit according to standard protocol and incubated for 6 hours with proteinase K. Three replicates were prepared using the Epicentre Stranded kit. cDNA samples were amplified for 12 PCR cycles and sequenced on an Illumina HiSeq 2500 instrument.

Study: Long-read sequencing of the Oxytricha trifallax macronuclear genome

Oxytricha trifallax's somatic genome is highly fragmented, with thousands of chromosomes averaging only 3.2 kbp. This makes it possible to capture many whole chromosomes in single PacBio sequencing reads. In this project we explore the benefits produce an updated version of the Oxytricha macronuclear genome including contigs that represent whole chromosome sequences derived from single reads.
