The goal was to input naturally occurring genetic variation (from 1000 Genomes individuals) into the mSTARR-seq MPRA to test for methylation-dependent genetic effects. Half of replicates were experimentally methylated and half were left unmethylated so that regulatory activity of input fragments (abundance of RNA relative to amount of input DNA) could be assessed in both contexts.
