Wild type strain H99 compared to independent sir2 deletion mutants grown to mid-log (0.6 OD600) in rich medium (YPD) shaking at 250rpm in baffled flasks. Overall design: Libraries were constructed from two biological replicates of wild type (Control), as well as two biological replicates of two independent sir2 delta clones.

Construction protocol: Total RNA was harvested from cell pellets by bead-beating (5 min, 30 sec on and off cooling on ice) followed by RNA extraction using the Qiagen RNEasy Mini Kit protocol. The samples were prepared using the Illumina TruSeq RNA library prep kit, starting with isolated mRNA since the organism was yeast. Approximately 1 ug of total RNA was used and polyA mRNA selected using the Illumina TruSeq RNA library prep kit. After multiplexing, the samples were pooled and sequenced on a HiSeq2500 in one lane, single read, 50 cycles.
