SRX13138005: RRBS of Octopus bimaculoides: 30 dpf whole-body hatchling
1 ILLUMINA (NextSeq 550) run: 28.9M spots, 8.5G bases, 3.6Gb downloads

Design: DNA quantity and quality was assesed by Qbit and Bioanalyer. 1000ng of DNA was used for library prep. Libraries were prepared according to Garrett-Bakelman et al., 2015 with some modifications. The adaptors used for multiplexing were purchased separately (Next Multiplex Methylated Adaptors - New England Biolabs). Libraries were size-selected by dual-step purification with Ampure XP Magnetic Beads (Beckman Coulter, Agencourt) to specifically select a region of fragments from 175 bp to 670 bp. Bisulfite conversion was performed with Lightning Methylation Kit (ZYMO Research) by following the manufacturers instructions. Libraries were amplified using KAPA HiFi HotStart Uracil+ Taq polymerase (Roche) and purified with Ampure XP Magnetic Beads (Beckman Coulter, Agencourt) before sequencing.
