SRX4925206: RNA-Seq of D. mojavensis from Cd. Obregon:adult male exposed to prickly pear diet

Design: Newly emerged adult males from prickly diet were washed twice with distilled water and placed into 1.5ml tubes, then rinsed twice with 1X PBS. All liquid was then removed, and flies were frozen (-70oC) until RNA extraction. Frozen samples were homogenized with Tri-Reagent using Teflon homogenizers and extracted using Direct-zolTM RNA MiniPrep extraction kit (Zymo Research) according to the manufacturer's protocol. Libraries were prepared with TruSeq RNA Sample Preparation Kit v2 (Illumina), selecting only polyA mRNAs and synthezing double stranded cDNAs to attach to the Illumina adapters. Library size and quality were measure by Bioanalyzer (Agilent Technologies) and sequenced in a 1 X 100 single-end read format on a HiSeqTM 2000 Sequencing System (Illumina).

Study: Transcriptional responses to seasonal dietary shifts in Drosophila mojavensis

Drosophila mojavensis normally breeds in necrotic columnar cactus, but they also feed and breed in Opuntia fruit (prickly pear) which serves as a seasonal resource. The prickly pear fruits are much different chemically from cacti, mainly in their free sugars and lipid content, raising the question of the effects of this seasonal shift on gene expression. We evaluated gene expression of emerging male and female adult flies exposed to prickly pear fruits or cacti using RNA-Seq.
