Design: Extracted DNA was repaired and end prepared by NEBNext FFPE DNA Repair Mix and NEBNext Ultra II End Repair/dA-tailing Module. Then adaptors were ligated on using Adapter Mix (AMX) and Quick T4 Ligase. Final product was loaded onto flow cell and sequenced.

Study: Nanopore Sequencing of Plasmodium Falciparum Strain SD01

Nanopore sequencing was performed on SD01, a clinical isolate of Plasmodium falciparum that originates in Sudan, in order to characterize better a genomic translocation between chromosomes 11 and 13, which results in the deletion of the pfhrp3 gene (an important gene in RDT diagnostics). The translocation involves a 15.3kb duplicated region between chromosomes 11 and 13 that contains rRNA loci; since this region is identical, long-range sequencing with Nanopore was needed in order to span this region in order to properly assemble this region on both chromosomes.
