Design: RNA extracted using Spectrum Plant Total RNA kit. RNA quality assessed. Real-time PCR with standardized 250 ng RNA. RNA samples checked on Caliper GX. 1.5 µg of RNA used as input of TruSeq Stranded mRNA Sample Prep kit for library preparation. mRNA fragmented and purified. Final libraries quantified and quality tested. Cluster generation with cBot and sequencing in paired-end mode on HiSeq2500.
