SRX1049907: transcriptome of TGMS SP2S at 16 Celsius degree
1 ILLUMINA (Illumina HiSeq 2000) run: 23M spots, 4.6G bases, 3Gb downloads

Design: Firstly, young floral buds from 4 treatments including FL (SP2F at 16°C), SL (SP2S at 16°C), FR (SP2F at 22°C), and SR (SP2S at 22°C) were collected, put into RNAlater, and stored at −80°C until further processing. Total RNAs were extracted using the Trizol® Reagent (Invitrogen, USA according to the manufacturer's instructions, and purified using TRK1001 Kit (LC Science, Houston, TX). The total RNA quantity and purity were analyszed by using of a Bioanalyzer 2100 and RNA 6000 Nano LabChip Kit (Agilent, CA, USA). We used 10 µg total RNA from each sample to create normalized cDNAs. The cDNAs were amplified according to the Illumina RNA-Seq protocol and sequenced on Illumina Hiseq2000 platform. Raw data generated from Solexa were trimmed to remove nonsense sequences including adaptor, low-quality reads, and very short sequences.
