Design: Wild-type Cryptococcus gattii R265 was pre-grown in YPD media (30 ºC for 24 h) and then inoculated in YNB media for a further period of growth (30 ºC for 18 h). YNB + 10 μM TPEN (for zinc deprivation) was inoculated with 1x107 cells/mL and incubated for additional 4 h at 30 ºC. Cells were collected by centrifugation (10,000 g for 10 min), and RNA was isolated by Trizol (Invitrogen) after cellular lysis via mortar and pestle. RNA integrity and concentration were assessed by electrophoresis on a 1% agarose gel and by fluorometry analysis using a Qubit fluorometer and a Quant-iT RNA assay kit according to the manufacturer’s instructions (Invitrogen). MRNA was purified from total RNA samples, processed and sequenced using Solexa technology on an Illumina Genome Analyzer GAII (Fasteris Life Sciences SA, Plan-les-Ouates, Switzerland).
