SRX15663492: RNA-seq of Neocallimastix frontalis strain EC30

Design: Total RNA was harvested from the growing strains isolated that had been maintained under anaerobic conditions initially isolated by a modified Hungate method. Enrichment of mRNA was performed by oligo(dT) beads. The mRNA is then fragmented randomly by fragmentation buffer, and cDNA synthesized from mRNA template and random hexamers primers. A custom second-strand synthesis buffer (Illumina), dNTPs, RNase H and DNA polymerase I are added to initiate the second-strand synthesis. After a series of terminal repairs, A ligation, and sequencing adaptor ligation, the double-stranded cDNA library is completed through size selection and PCR enrichment.
