SRX8816737: Exome Capture of Triticum aestivum
1 ILLUMINA (Illumina NovaSeq 6000) run: 109.6M spots, 33.1G bases, 9.4Gb downloads

Design: Genomic DNA was isolated from leaf tissue and sent to Arbor Biosciences (Michigan, USA) for capture. After DNA fragmentation and library preparation target enrichments were performed using 750 ng of each library and the Arbor Wheat Exome Beta probe set (as described in the standard myBaits 4.0 manual). Following cleanup libraries were sequences on NovaSeq S4 (Illumina) instrument.

Study: Towards the identification of the ph2 gene in hexaploid wheat

In spite of being hexaploid, recombination in wheat only occurs between homologous chromosomes whereas recombination between homoeologous chromosomes is suppressed. The two main loci controlling homoeologous recombination are located on chromosome-arms 5BL and 3DS, named Ph1 and Ph2 respectively. While Ph1 has been characterized, the causative gene sequence for Ph2 is yet to be determined. The present study aims at the identification of Ph2 gene candidates by RNASeq and exome capture of DNA from the ph2a deletion and the ph2b EMS-derived mutants.
