SRX7196006: Whole genome sequencing of Spiroplasma citri strain BLH-13

Design: Using LD8 medium, S. citri strain BLH-13 was isolated from beet leafhopper collected from parsley in 2010 near Mettler, CA and maintained in culture. For sequencing, S. citri strain BLH-13 was triple-cloned and cultured in LD8 medium to obtain enough DNA for sequencing. Total genomic DNA was extracted using CTAB method and library was prepared for sequencing using PacBio SMRTbell Express Template Prep Kit. Whole-genome sequencing was performed on the PacBio RS II platform. Reads were filtered and assembled using SMRT Analysis software from PacBio with default settings.

Spiroplasma citri is a bacterium that causes stubborn disease of citrus and is transmitted by leafhoppers in a circulative and persistent manner. Due to limited sequence data on S. citri, several strains of bacterium were isolated from various hosts and locations in California. DNA from S. citri was extracted from a pure culture in LD8 and subjected to PacBio sequencing. The genomes developed extend the sequence database of S. citri and allow for comparative genomics analyses.
