SRX4221801: RNA-seq of pooled gill tissue samples from 12 walleye

Design: Total RNA was extracted from gill tissues using RNeasy Plus Mini Kits (Qiagen). Concentration and integrity of the RNA were assessed using a Nanodrop One Spectrophotometer, a Qubit, and gel electrophoresis. Two random samples were chosen from six sampling locations (n = 12 samples) and equal amounts of total RNA (2000ng) were pooled. The quality of the pooled sampled was assessed using a Bioanalyzer (Agilent) and had a RIN of 8.6. The pooled sample was used for a stranded mRNA isolation library preparation using a NEBnext library prep kit. Sequencing was conducted to produce 100 paired-end reads over one Illumina HiSeq 4000 lane.

Study: Generating a Reference Transcriptome for Walleye

Walleye (Sander vitreus) are an important commercial and recreational fish species in North America. Lake Winnipeg, Manitoba, supports the second largest freshwater fishery in Canada, with walleye being a focal species. In 2017, wild-caught walleye from six different locations in the Lake Winnipeg and Lake Manitoba basins were collected and sampled for gill tissue. One of the goals of a larger project examining movement patterns in Lake Winnipeg is to develop molecular tools for examining the effects of environmental stressors on walleye in Manitoba. We used gill tissue samples from individuals from the different sampling locations to develop a reference transcriptome for future studies examining the effects of environmental stressors on this species.
