SRX1880621: Whole transcriptome sequencing of Pistacia vera
1 ILLUMINA (Illumina HiSeq 2000) run: 197.3M spots, 39.5G bases, 16.1Gb downloads

Design: High-quality total RNA was extracted from leaf, stem and root of pistachio plants and mixed an equal amount to make a single pool. Following mRNA enrichment using magnetic oligo (dT) beads, library preparation from the pooled RNA sample was performed as outlined in Illumina’s TruSeq Stranded mRNA Sample Prep Kit. The cDNA library was sequenced on the one lane of Illumina Hiseq 2000 platform as 2 × 100 run.

we have performed transcriptome sequencing of leaf, stem, and root of two pistachio cultivars (Pistacia vera L. Sarakhs and P. vera L. Ghazvini) under salinity and control conditions on one lane of Illumina Hiseq 2000 to produce a robust pooled transcriptome assembly of P. vera. Our study, as the first report on the transcriptome sequencing of pistachio, will not only provide plenty of informative data, but also paves the way for functional and comparative genomic studies on this major tree and other woody species to decipher the potential abiotic stress response mechanisms.
