Purpose: The goals of this study are to compare The difference of RNA expression between Pumpkins seed coat and hulled-less Pumpkins seed coat. Methods:Using Trizol kit, total RNA was extracted from seed coats of squash with and without shells under the conditions of 5 DAP, 10 DAP and 20 DAP. Then, 1 µ g RNA was extracted from each sample using the Illumina Hiseq 2000 platform for sequencing. Overall design: Total RNA was extracted from seed coats of hulled and hull-less pumpkins at 5 DAP, 10 DAP, and 20 DAP using a Trizol kit (Invitrogen, USA). Then, 1 µg RNA from each sample was taken for sequencing using an Illumina HiSeq 2000 platform. Three biological replicates were performed for each stage. Transcriptome analysis was performed as described by Trapnell et al.
