SRX24461503: RNA-seq of Magnolia lotungensis: bark, leaf and flower

Design: For the RNA-sequencing of Magnolia lotungensis, we extracted total RNA from bark, leaves, and flowers using the Qiagen RNeasy Plant Mini Kit. Following quality assessment with a NanoDrop 2000 and Agilent 2100 Bioanalyzer, we constructed RNA-seq libraries with the NEBNext Ultra RNA Library Prep Kit for Illumina. The mRNA was enriched, fragmented, and converted to cDNA, followed by end-repair, A-tailing, and adapter ligation. The libraries were size-selected, amplified by PCR, and checked for quality before sequencing on the Illumina HiSeq 2500 platform with a 150 bp paired-end run.
