SRX534461: Mitogenomic sequences effectively recover relationships within Nymphalidae butterflies

Design: Thirty entire mitogenomes were amplified by two fragments, one for ~6.5kb in length and the other of ~9.5kb in length. PCR products were checked using 1-2% agarose gel, and each product was purified using commercial purification kit. The two long PCR products were mixed in equimolar concentrations, and then sheared into 200-1000 bp. Each sample was ligated with Roche adaptors and barcode-tag. Total of 30 tagged-samples were mixed together and sequenced using the Roche 454 GS Junior System. Mitogenomic phylogenies have revealed well supported relationships for many groups of eukaryotes, including Lepidoptera. However, these data is restricted to 7 of 43 known superfamilies, and it remains challenging to recover large numbers of mitogenomes at once. Therefore, we present a procedure to obtain a large number of mitogenomes via the Roche 454 GS junior system.
