SRX9863259: Lake Constance metatranscriptome study
1 ILLUMINA (NextSeq 500) run: 148.5M spots, 41.6G bases, 15.5Gb downloads

Design: For metatranscriptome sequencing, messenger RNA was enriched by depleting ribosomal RNA with the Ribo-off rRNA Depletion Kit (Bacteria; Vazyme, Nanjing, China) or the Ribo-Zero Bacteria Kit (Illumina) to gain higher mRNA concentrations from the samples. The mRNA library was prepared with the TruSeq Stranded mRNA Library Prep (Illumina) and sequenced on a NextSeq500 sequencer with the NextSeq 500/550 Mid Output Kit v2.5.

Lake Constance is the largest lake in Germany with a volume of 48 km3 and a maximum depth of 250 m. It is oligotrophic and has a permanently oxygenated hypolimnion throughout the year and serves as a drinking water reservoir for more than 5 million people. Samples were taken in 85m depth and was pre-filtered through a 70 and 30 micron nylon mesh and filtered on board onto 5 and 0.2 micron polycarbonate filters. DNA was extracted from 0.2 micron filters to obtain genomic DNA for metagenome sequencing.
