1 ILLUMINA (Illumina HiSeq 4000) run: 18.2M spots, 5.5G bases, 2.2Gb downloads

Design: Library preparation was performed using IlluminaÕs NexteraXT library prep kit. 1 ng of total genomic DNA was used as starting material and tagmented with Illumina adapters and unqiue 8 bp dual indices. Following library prep, libraries were checked on an Agilent Bioanalyzer 2100 with a High Sensitivity DNA analysis kit. Subsequently, libraries were normalized and multiplexed using the molarity between 250-400 bp. The multiplexed libraries were then purified on a 2% agarose gel and DNA was size selected between 250-400 bp for extraction using a Qiaquck gel extraction kit (QIAGEN) for sequencing. The purified and size selected libraries were then sent for sequencing on an Illumina Hiseq platform using 150 bp paired end chemistry. Data were then demulitplexed by sample and raw data were sent for analysis.
