We examined a combination of sample handling choices (sample storage, extraction kit and 16S amplification) for swab material using an even mock community (HM-280). In addition, blank controls were used to monitor the level of laboratory and kit-specific contamination. By examining a combination of effects, we aim to contribute to the ongoing discussion regarding standardization of sample handling for amplicon and metagenomic sequencing.

Sample: A cell mixture of 22 different bacterial strains at a concentration of 1x10^8 cells/mL of each organism (Microbial mock community HM-280) in phosphate buffer saline (PBS) from the Biodefense and Emerging Infectious Research (BEI) Resources of the American Type Culture Collection (ATCC) (Manassas, USA)
