Design: The pADL-10b plasmid was isolated from E. coli samples that were used for phage expression. Primers specific to the library region of the pADL-10b plasmid were then used to create amplicons of the library region. Two subsequent PCR steps were then performed to attach Illumina adapters and indices that allowed for Illumina sequencing of the amplicons.

In this study, we aim to develop amber obligate phage libraries that will allow for efficient incorporation of unnatural amino acids into phage libraries via amber suppression. Analysis of this amber obligate library was done through Illumina sequencing of amplicons of the phage library on pIII.
