Design: TCR repertoire library was prepared by using multiple specific primers for V gene segments of TCR and a universal reverse primer specific to the constant region of TCR. In the rst round of amplication, cDNA was mixed with 23 ILMN_TRBV mixed primers, 1 specic ILMN_TRBC primer as wellas the 1e7 copies of barcoded plasmid template mixture. A second round PCR was done to add the specific P5 and P78 llumina index primers.The purified product was sequenced on an Illumina HiSeq Xten instrument.
