Examples of agonist and antagonist concentration-response curves in G_αi-coupled receptors. Panel A. The agonist and antagonist responses are plotted as the HTRF ratio (Em665/Em615×10⁴) along with the cAMP standard curve. Note that both types of response produce signals that are within the linear portion of the standard curve. Panel B. The agonist and antagonist responses are plotted as the cAMP level to determine the EC₅₀ and IC₅₀ values, respectively. Data are analyzed using four-parameter logistic regression.
