Title A Clock-Driven Neural Network Critical for Arousal

Summary Purpose: scRNA-Seq was performed to profile hypothalamic mWake+ cells of male at 7 weeks old
 
Overall design The hypothalamus of mWake-tdTomato-Cre mice (male, 7 weeks old) were dissociated using modified Act-Seq protocol and flow-sorted to enrich tdTomato+ cells. A small aliquot of flow-sorted cells were labelled with DAPI and counted under the microscope (99% tdTomato+ cells). Cells were loaded into V3.0 10x Genomics Chromium Single Cell system, and 3 libraries (triplicates) were sequenced on Illumina NextSeq 500 with ~150 million reads per library. Sequencing results were processed with 10x Genomics pipeline. Seurat V3 was used in conjunction with Monocle3 to perform downstream analysis.
