The cells were cultured in minimum essential alpha-medium (αMEM) supplemented with 20mM HEPES, 10% heat-inactivated fetal bovine serum (FBS), 2mM L-glutamine and 100 units/ml penicillin and 100 μg/ml streptomycin (all from Gibco, Paisley, UK). Same serum lot was used throughout the study. The medium was renewed twice a week and the cells were harvested when 70-80% confluent and plated further at density of 1000 cells/cm2. Cells were cultured until senescence was reached.

RNA was extracted using the Qiagen AllPrep DNA/RNA Mini Kit (Cat. no. 80204, Qiagen, CA, USA) and a Qiagen supplementary protocol “Purification of total RNA containing miRNA from animal cells using the RNeasy Plus Mini Kit (cat. no. 74134).
