Extraction protocol RNA was harvested using Trizol (Takara). 800 ug of total RNA was used for the construction of sequencing libraries.
The extracted mRNA is enriched using mRNA Capture Beads. After purification with beads, the mRNA is fragmented using high temperatures. The fragmented mRNA is then used as a template to synthesize the first strand of cDNA in a reverse transcription enzyme mixture system. While synthesizing the second strand of cDNA, end repair and A-tailing are completed. Next, adapters are ligated, and Hieff NGSĀ® DNA Selection Beads are used for purification to select target fragments.
