Hearts were excised and continiousy perfused with enzymatic buffer solution containing 2 mg/mL collagenase IV (LS004188, Worthington Biochem) and 1 mg/mL dispase II (04942078001, Roche). Single cell suspension were then stained with antibodies and with Sytox™ Green (S34860, Invitrogen) and Vybrant™ DyeCycle™ (V10273, Invitrogen) and verapamil and sorted for live, metabolically active and nucleated cells using FACSAria™ III cell sorter (B.D.) Libraries were prepared using according to the 10x Genomics chemistry 3` v2 kit and sequenced in an Illumina HiSeq instrument. Analysis was performed using 10X Genomics pipeline.
