Extraction protocol Single-cell suspensions at 1 × 105 cells/mL in concentration in PBS were prepared and loaded onto microfluidic devices.
Libraries were constructed according to Singleron GEXSCOPE protocol by GEXSCOPE Single-Cell RNA Library Kit (Singleron Biotechnologies), which included cell lysis, mRNA trapping, labeling cells (barcode) and mRNA (UMI), reverse transcription mRNA into cDNA and amplification, and finally fragment cDNA.

Read 1: contains the 24bp GEXSCOPE barcode and the 12bp UMI (bases 25-36). Read 2 contains the cDNA information.
