Growth protocol |
UM53 and ASPC1 cell lines were grown in RPMI media supplemented with 10% FBS. The cellswere maintained in 37°C with 5% CO2 and tested to be mycoplasma-free.

Genomic DNA exptration was carried out using the Wizard Genomic DNA purification kit (Promega, A1120) according to the manufacturer's protocol A two step PCR was performed to enrich guide RNA regions in the genome and to amplify the guide RNAs with Illumina TruSeq adapters with i5 and i7 indices.
