Status Public on Jan 24, 2024
Title UM53-TKO-MRTX-500-P8
Sample type SRA

Source name UM53
Organism Homo sapiens
Characteristics cell line: UM53
treatment: MRTX849, 500nM
Treatment protocol The positive and negative selections of the guide sequences in ASPC1 cells were carried out using 0.1% DMSO and MRTX1133 (6.25 nM). For the UM53 the selectio was performed using 0.1% DMSO and MRTX849 (500 nM)
Growth protocol UM53 and ASPC1 cell lines were grown in RPMI media supplemented with 10% FBS. The cellswere maintained in 37°C with 5% CO2 and tested to be mycoplasma-free.
Extracted molecule other
Extraction protocol Genomic DNA exptration was carried out using the Wizard Genomic DNA purification kit (Promega, A1120) according to the manufacturer's protocol
A two step PCR was performed to enrich guide RNA regions in the genome and to amplify the guide RNAs with Illumina TruSeq adapters with i5 and i7 indices.
