About 3000 HSCs of BMs from Nynrinfl/flTie2-Cre or Nynrinfl/fl mice were sorted by FACS,and extraction and Purification of total RNA using Trizol. 5 ng cDNA was used for the construction of sequencing libraries. The construct of the whole cDNA library for low number HSCs from from Nynrin cko mice was performed as previously described (Chen et al., 2017, PMID: 28207000).
