Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using Leucocyte kit (Norgen, Canada) as per manufacturer's instructions
Label |
biotin
Label protocol |
Single-stranded cDNA was generated from the amplified cRNA with the WT Plus cDNA Synthesis Kit (Affymetrix) and then fragmented and labeled with the WT Terminal Labeling Kit (Affymetrix).
Hybridization protocol |
Whole-genome gene expression profiling was performed using the Clariom S human gene expression array (Affymetrix) with Hybridization, Wash, and Stain Kit (Affymetrix) following manufacturer's instructions
Scan protocol |
Clariom S chips were scanned with the GeneChip Scanner 3000 7G (Affymetrix)
Data processing |
Raw data was imported to Affymetrix software Transcriptome Analysis Console (TAC) and quality control (QC) was performed as manufacturer's instructions (PCA, labeling controls, hybridization controls, Pos vs. Neg AUC, and Signal Box Plot). Normalization was performed with the Robust Multi-chip Analysis (RMA) approach with default analysis settings.
