Single cellsof retina tissueswere isolated by using Neuronal Isolation Enzyme (with papain)(Thermo Fisher,#88285)
The libraries were constructedusing the Chromium platform and Chromium Single Cell 3′ v2 chemistry. Briefly, cellular suspensions were loaded on a Chromium Single Cell Instrument (10x Genomics, Pleasanton, CA) forGEMs Generation and Barcoding.Single cell RNA-Seq libraries were prepared using the Chromium Single Cell Library Kit (10x Genomics, Pleasanton, CA).

Illumina BCL files were converted to fastq files by using 10X Genomics Cell Ranger pipeline mkfastq function
Cellranger count function was used to generate Gene-Barcode matrices from the FASTQ files, which uses the STAR algorithm to map high-quality reads to the human reference genome (mm9), then performs UMI counting.
