CWR22Rv1-AR-EK cells were transfected with 25 nm siRNA for 48 hours then treated with either DMSO or 1 µM NU7441, NU5455 or AZD7648 for a further 24 hours, CWR22Rv1 cells were reverse transfected with 25 nm siRNA for 72 hours

CWR22Rv1-AR-EK and CWR22Rv1 cells were maintained in RPMI-1640 supplemented with 10% fetal bovine serum (FBS) and penicillin-streptomycin antibiotics in humidified atmosphere with 5% CO2 at 37 °C
