Treatment protocol Pregnant mice were treated daily by oral gavage with 30mg/kg of L21 or Iso-L21 from embryonic day 5.5 to E13.5.

Extraction protocol E13.5 frozen embryonic hearts were placed in lysis buffer and homogenized using a cordless Pellet pestle (Sigma-Aldrich) followed by RNA extraction using RNAeasy micro kit (Qiagen)
polyA enriched libraries usingNEBNext®Ultra™II Directional RNA Library Prep Kit for Illumina according to manufacturer’s instructions. An average 25 million pair end reads per library (PE100) were generated on a NovaSeq6000 (Illumina).
