Extraction protocol |
Transfer fresh tissue to a culture dish containing HBSS buffer and wash it three time. Afterwards, remove the excess buffer and transfer the tissue to a 1.5mL low-adsorption centrifuge tube. Add 200 μL of Tissue Dissociation Mix, and use sterile ophthalmic scissors to cut the tissue to minced form. After adding 1 mL of Tissue Dissociation Mix, the minced tissue homogenate was transferred to a 15 mL low-adsorption centrifuge tube for tissue dissociation. The dissociated cell suspension was filtered through a 40um filter. And the Single-cell suspensions were prepared at a concentration of 1 × 105 cells/mL in PBS (HyClone). Single-cell suspensions were loaded onto microfluidic devices and scRNA-seq libraries were constructed according to Singleron GEXSCOPE® protocol by GEXSCOPE® Single-Cell RNA Library Kit (Singleron Biotechnologies). Individual libraries were diluted to 4nM and pooled for sequencing. Pools were sequenced on Illumina NovaSeq with 150 bp paired end reads.
