We carried out expression profiling experiments in which gene expression in logarithmically growing cultures of HDAC mutant cells was compared to that in wt controls. Wt, hos2D, clr6-1, clr3D and sir2D cells were grown to midlogarithmic phase (5*10power6 cells/ml) in rich medium, and RNA was extracted and subjected to cDNA expression profiling analysis using the established protocols (Xue et al, 2004).To investigate the roles of HDACs at heterochromatic genes we also analyzed Swi6, the fission yeast heterochromatin protein 1 (HP1) homolog, by expression profiling of swi6D and Swi6 over expression (pSwi6) strains.
