Sample GSM6467387 Query DataSets for GSM6467387
Status Public on Oct 07, 2024
Title BLaER1 Mo internalizing Mito- RBCs
Sample type SRA

Source name Cell Line
Organism Homo sapiens
Characteristics tissue: Cell Line
cell line: BLaER1 cells
genotype: WT
treatment: culture with Mito- RBCs
Treatment protocol Cells were co-cultured with Mito- or Mito+ RBCs for 18 hr
Growth protocol Human Mo were sorted from blood
Extracted molecule total RNA
Extraction protocol Total RNA was isolated from cell lysates using the RNeasy Micro Kit (QIAGEN)
Poly(A)-enriched next-generation sequencing library construction was performed using the KAPA mRNA Hyper Prep Kit (KAPA Biosystems) with 100 ng of input total RNA and 15 amplification cycles
The quality and quantity of the individual libraries was assessed using the High Sensitivity DNA Kit (Agilent). The libraries were equimolar pooled, and an additional 1X bead clean-up was used to remove adaptor dimer. The pooled libraries were quantitated using the KAPA Library Quantification Kit, Universal (KAPA Biosystems) and sequenced on an Illumina NextSeq 500 with paired-end 75-base-pair (bp) read lengths.

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NextSeq 500

Data processing Adapter and quality trimming of sequencing reads were performed using TrimGalore
HISAT2 was used for aligning sequncing reads against the human genome GRCh38
Gene quantification was done using feature-counts
Assembly: GRCh38
Supplementary files format and content: tab-delimited file with raw gene counts for each sample
