Treatment protocol |
Irradiated cells were irradiated with 2.0 Gy of gamma radiation. Control cells were incubated in medium from non-irraidated cells 2 h after irradiation. Bystander cells were incubated with medium from cells irradiated with 2.0 Gy, 2 h after irradiation. Extraction of RNA was done 4, 6 and 8 h after irradiation.

Growth protocol |
The cells were grown in DMEM supplied with 1% L-Glutamine and Penicillin/ Streptomycin and 20% FBS and kept at 37°C and 5% CO2.

Total RNA from 4 independent experiments was isolated from cells irradiated with 2.0 Gy and bystander fibroblasts 4, 8 and 26 h after irradiation using Pure Link RNA mini kit (Invitrogen, Carlsbad, CA, USA) according to the manufacturer’s instructions.
