Hearts were dissected and dissociated into single cell suspensions using the Neonatal Heart Dissociation kit (Miltenyi Biotech) and gentleMACS Octo Dissociator (Miltenyi Biotech) with gentle MACS C tubes and were subsequently methanol-fixed and stored at -80C. libraries were prepared with the 10x Chromium 3’ mRNA-Seq kit version 2. The cells were QC’d with trypan blue viability dye and quantified using the EVE automated cell counter. Approximately 2000 – 3000 cells were loaded onto the 10x and prepared according to manufacturer’s instructions.

Cellranger-2.1.1 was used to deconvolve, align and quantify the per-cell transcriptomes. Reads were aligned to mouse mm10-1.2.0 (obtained from 10x genomics) Genome_build: mouse mm10 (mm10-1.2.0 from 10x Genomics) Supplementary_files_format_and_content: tar file containing 10x counts matrix, features and barcode data from filtered cell ranger outputs
