Treatment protocol The mice were administered with Ad-Inpolin or Ad-GFP (1×109 plaque-forming units) through the tail vein and sacrificed at 12th day of administration.
Growth protocol Male ob/ob mice aged 8-12 weeks were purchased from GemPharmatech Company (Nanjing, China).

Data processing Paired-end reads were harvested from Illumina NovaSeq 6000 sequencer, and were quality controlled by Q30.
After 3’ adaptor-trimming and low quality reads removing by cutadapt software (v1.9.3), the high quality clean reads were aligned to the mouse reference genome (UCSC MM10) with hisat2 software (v2.0.4).
Then, guided by the Ensembl gtf gene annotation file, cuffdiff software (part of cufflinks) was used to get the gene level FPKM as the expression profiles of mRNA.
Fold change>=2.0, that is, log(FC)>=1.0, p-value<=0.05, and FPKM value>=0.1 in at least one (or a group of) samples as the threshold for differential screening.
