strain: rap1-2 ts mutant
|
Treatment protocol |
Standard chromatin/Mnase prep, digesting to mostly mononucleosomal DNA
|
Growth protocol |
Wild type and matched mutant yeast were grown at 25 C, shifted for to 37 C and grown at 37 C for one hour before harvesting. Note third Rap1 samples (Tsui samples) were grown 2 hrs at 37 C.

Extracted molecule |
genomic DNA |
Extraction protocol |
Phenol/chloroform after proteinase K and cross link reversal; DNA was fragmented with DNase prior to labeling
|
Label |

|
Label protocol |
Labeled with biotinylated ddATP using terminal deoxynucleotidyl transferase

Description |
nucleosomal DNA Rap1 ts mutant against genomic DNA
|
Data processing |
The data was analyzed using TAS v1.1.02, and normalized using built-in quantile normalization and probe-level analysis with both perfect match and mismatch (PM/MM) probes and run with a bandwidth of 30. Text versions of the processed data (BAR) files are also provided on the Series record. Three Abf1 ts against wild type, and three Rap1 against WT; 2-sided t-test for pvalues
